Abstract

TMV (Tobacco mosaic virus) is a virus that causes mosaic disease in tobacco. Symptoms of the disease caused by TMV appear in the leaves which change color to yellowish green in an irregular pattern. The light colored parts do not develop as fast as the normal green parts so the leaves wrinkle and twist. This study aims to (1) show how mechanical transmission of TMV occurs; (2) to show how long TMV survives on dry leaves; and (3) to show the symptoms of TMV infection in tobacco. The tools used in this practicum are mortar, pistil, 10 ml measuring cup, plastic, support rod, spray bottle, camera, and writing tools. Meanwhile, the materials used are inoculum sources in the form of fresh and dry tobacco leaves (dried for 1 month) infected with TMV, healthy tobacco plants, phosphate buffer, distilled water and carborundum. The results of incubation on test tobacco plants inoculated with TMV with each treatment showed that fresh leaf inoculum treatment with wounds showed symptoms of attack in the fastest latent period, namely day five. This result is in line with the greater number of infected leaves, in the first week the tobacco leaves were infected compared to other treatments. Symptoms caused by wound treatment from both fresh and dry inoculum sources showed symptoms that were more severe than others
